Figure 23 illustrates the effectiveness of one, two, and four fractions of radiation. Since in each case full recovery has occurred between doses, the shoulder portion of each curve has reappeared. Because of the reappearance of the shoulder between fractions, a larger total dose is required to reduce the surviving fraction to a given number; ie, the greater the number of fractions, the larger the total dose required for a given biologic effect. One of the earliest and best known studies attempting to correlate radiation dose with time is that of Strandquist [ 12].

Figure 2-2 illustrates the results of those experiments. Within 2 hr, the cells had reached their pre irradiation sensitivity; however, following this, the cells were more sensitive to radiation and this sensitivity reached a peak approximately 4 hr following the first 2 4 6 8 1. Repair of sublethal damage occurs quickly and is usually complete within 2 hr of irradiation. 2. For a short time, cells become synchronized following this first dose of radiation. 3. The sensitivity of cells to a second dose of radiation will depend on the phase of the cell cycle at the time the radiation is given.

When the irradiated cells finally progress into S phase, however, they usually synthesize DNA at normal rates. The same observation has been made for cells irradiated during the stage of DNA synthesis. For example, a dose of about 500 rads delivered to synchronous HeLa cell populations in S phase reduced DNA synthesis by 50% with prolongation of S phase [72]. Very little effect on DNA synthesis occurred after irradiation in other phases of the cell cycle. Overall, total DNA synthesis was essentially normal [72].
